BDGP P1 Vectors

Two types of P1 vectors

The P1 mapping library consists of clones generated in two related P1 cloning vectors. 3,840 clones were inserted into the vector known as pNS583tet14Ad10 (Smoller et al. 1991) and 5,376 clones were from the ligation mixture using the vector called pSacBII (Kimmerly et al. 1996). Both vectors contain genes that allow resistance to the antibiotic kanamycin, a plasmid origin of replication that provides for a single copy, a loxP site (the cis-acting site specific recognition signal for the P1 recombinase), and a gene that allows resistance to tetracycline. Inserts cloned in the vector pNS583tet14Ad10 have been introduced into the BamHI site in the gene allowing tetracycline resistance. A sacBII cassette was introduced by Sternberg (1990) as a means of selecting for clones with inserts. The cloned inserts in this vector system are introduced between SP6 and T7 RNA polymerase promoters that are positioned of the Bacillus subtilis genes found in the SacBII cassette. The sequences between the loxP sites (containing the Ad10 sequences) are deleted during library construction and thus are not present when the clones are mapped and eventually sequenced.
